The CVS site has been the site of a flourmill for centuries - the presence of a Twyford Mill apparently being recorded in the Domesday Book. At the beginning of the 20th Century activities on the site included iron-ore extraction and smelting to the north and east of the Mill, a brickworks operated at Twyford Wharf and flour milling. These industries were all well placed to take advantage of the Great Western Railway and Oxford Canal which cut through the valley.

Twyford Mill and the surrounding land was acquired by the Cherry family, the current proprietors, in 1935 and since that time a variety of activities have operated on site including dairy farming,saw milling, seed production, farm requisites, electrical goods retailing and poultry rearing. In 1936, Ken Cherry set up Twyford Mill Ltd which was located in the Old Twyford Mill. This company became Twyford Seeds in 1946 when it moved to the old Beaverbrook factory in Adderbury. Twyford Seeds and the Adderbury site were sold to J Bibby Ltd in 1952. From 1957 to approximately 1970 a poultry rearing and laying hen facility operated from the site. At its peak, this operation had 30,000 laying birds.

Since 1970, Cherwell Valley Silos Limited has run its transport and animal feed ingredients businesses from the site.
